Early in her career, she appeared in Scream of the Shalka, a Doctor Who webcast, providing both the voice and likeness for the character Alison Cheney, and later portrayed Liz Ten (Queen Elizabeth X) in the 2010 BBC episodes “The Beast Below” and “The Pandorica Opens.” 

She also appeared in the BAFTA-nominated Clocking Off as Jenny, and played Tulip Jones in Stormbreaker (2006), Nancy in the 2007 adaptation of Oliver Twist, and the Wachati Princess in Ace Ventura: When Nature Calls (1995). In 1998, she was considered for the lead in B. Monkey, but the role was blocked by the producer.

Okonedo earned an Academy Award nomination for Best Supporting Actress for Hotel Rwanda (2004) and a Golden Globe nomination for her role in Tsunami: The Aftermath (2006). She appeared in The Secret Life of Bees (2008), Skin (2009), and portrayed Winnie Mandela in the BBC drama Mrs. Mandela (2010).

Her stage work has been widely acclaimed. She starred as Ruth Younger in the 2014 Broadway revival of A Raisin in the Sun, winning the Tony Award for Best Featured Actress in a Play.
